CARDIOFISH系統(tǒng)是一款最新開發(fā)的檢測成年斑馬魚心臟功能的產(chǎn)品,該系統(tǒng)可以有效地用于測試藥理學(xué)、安全藥理學(xué)(safety pharmacology)和毒理學(xué) (toxicology)等藥物分子的效果和副作用, 它也是很適合心律失常(arrhythmias )和regener-ation心臟肌肉能力的基礎(chǔ)研究。
CardioFish系統(tǒng)主要用于斑馬魚心率監(jiān)測,系統(tǒng)包括三個單元:
1. 機(jī)械裝置
2. 低噪音寬頻模擬放大器
3. 數(shù)據(jù)可視化 (visualization),存檔(archiving)和-評測(evaluation)硬件/軟件
特殊的法拉第籠(The Faraday cage)無振動物體箱固定有系統(tǒng)兩個主要部件(顯微操作儀和寬頻放大器),它確保完全屏蔽電氣和磁場的干擾,還附帶有一個小魚缸和兩個顯微操作儀,顯微操作儀可以很容易地水平調(diào)整和移動電極固定器,電極固定器可安裝我們的不銹鋼電極或標(biāo)準(zhǔn)的玻璃電極。
測量數(shù)據(jù)(MEASURING DATABASE)
采集和分析框架是源自經(jīng)典的并已驗證的記錄和分析系統(tǒng)(Cardiosys),該系統(tǒng)已用于心臟 (cardiac R&D)研發(fā)15年。軟件記錄數(shù)據(jù)和處理是通過其中的Cardiofish Z-01軟件來管理的,它有一個內(nèi)置的數(shù)據(jù)庫架構(gòu),所有測量結(jié)果自動存儲,數(shù)據(jù)庫可以通過快捷按鈕打開。

1. 斑馬魚心電記錄(ECG recordings)可通過監(jiān)視畫面上的快捷按鈕和滾動菜單進(jìn)行查看,心電曲線(ECG curve)可應(yīng)用可視化濾波器或出現(xiàn)的視覺參數(shù)加以改變(如:amplifica-tion、速度), 測量值注釋可以被記錄,各個數(shù)據(jù)記錄處理可以單獨(dú)的被啟動或停止。
2. 采樣頻率可以在0.5和5 kHz之間調(diào)整,同時提供了一個合適的低通轉(zhuǎn)角頻率設(shè)置(Nyqvist頻率),可以確??焖俚脑敿?xì)特征信號(QRS)保存。
3. 斑馬魚心電(ECG)信號顯示可通過設(shè)置時間窗口寬度和放大顯示進(jìn)行調(diào)整,最優(yōu)化的過濾效果(50或60Hz陷波濾波器,高通基線過濾器(刪除breath-related基線振蕩)和低通高頻帶阻濾波器)是由可視化疊加和過濾信號輔助的,用戶可以循著記錄點進(jìn)行時間分類標(biāo)注,很容易的截取感興趣的部分進(jìn)行脫機(jī)制作分析。
離線分析(OFFLINE ANALYZATIONS )
記錄的ECG心電圖信號可以在測量后進(jìn)行正確評估或以后任何時候都可以評估,軟件提供了五種不同的可能性,最典型的(ECG)心電圖參數(shù)可以快速計算,以及更復(fù)雜的功能,以及更廣泛的選擇:
1. 單光標(biāo)分析(One cursor analysis): 一個應(yīng)用于收集振幅,在用戶心電圖(ECG)信號上定義光標(biāo)位置和HR值被收集到一個帶統(tǒng)計功能的圖表里。

4. 心電圖QT分析(ECG QT analysis): calculating an eligible QT or QTc value, as well as QT variability (QTv) as a function of RR or HR, in case of at least 32 chosen cycles. Criteria of reliability of the measurements are also evaluated.
5. 心臟頻率變化(HRV)分析(Heart frequency variation (HRV) analysis): 心臟頻率的變化是隨時間和頻率分析而變化的, 也提供有正常化得RR。

該斑馬魚測試系統(tǒng)主要用以建立一個高通量的能夠快速和經(jīng)濟(jì)有效的在很短的時間內(nèi)研究大量分子心臟影響方面的系統(tǒng)。STUDYING THE EFFECTS OF FLUORIDE EXPOSURE ON CARDIAC FUNCTION IN ZEBRAFISH (DANIO RERIO)

In the last few decades the effects of fluoride have been well-studied in many organisms. In mammalian models it was found that long-term fluoride exposure changes the ECG profile, prolongation of the P-Q and Q-T intervals were detected. Our aim was to study the effects of chronic fluoride exposure on adult zebrafish and to find out if these effects are similar to those found in mammals.
Fish were treated with 50 ppm, 100 ppm and 200 ppm NaF prior to the ECG measurement. Solutions were changed twice a week during the 3 months study. ECG was measured monthly, for data acquisition and analysis the newly developed zebrafish ECG measurement system (EXPERIMETRIA Ltd.) and the ADVANCE HAEMOSYS haemodynamic software were used. To complete our results, short- term effects of fluoride to heart rate were studied on zebrafish embryos. Embryos were treated with 500 ppm, 400 ppm, 200 ppm, 100 ppm, 50 ppm NaF solutions for 3 days. Embryonic heart rate (HR) was analysed on the basis of 30 second video records.
Results correlated well with the mammalian models. In the third month, significant Q-T prolongation (p<0,05) was found in the 100 ppm (414,83ms) group. The Q-T interval increased in the 3rd month 200 ppm (473,06group too, but it wasnt significant. We also found significant (p<0,05) HR decrease in the 3rd month 200 ppm (101,041/min) group. Embryonic HR increased significantly (p<0,01) on second and third days in the 500 ppm groups.
